Film ini berawal pada suatu malam Peter Parker sedang bersantai di sarang laba-laba nya bersama pacarnya Mary Jane, suatu ketika jatuh sebuah meteor yang membawa ma hluk asing yang bersifat parasit, mahluk asing tersebut akhrinya ikut bersama Peter Parker, dan pada suatu malam mahluk asing tersebut merasuki tubuh Peter Parker. Maka, Peter alias Spiderman kemudian berubah menjadi agresif dan percaya diri. Bahkan cenderung jahat dalam menghadapi segala persoalan yang dihadapi. Termasuk menghadapi Harry Osborn (James Franco), sahabat Peter yang sekaligus anak dari musuh besarnya, Green Goblin, yang telah meninggal. Harry (diakhir film Spiderman 2) mengetahui identitas Spiderman sebenarnya. Sejak saat itu bertekad untuk menuntut balas kepada Spiderman yang diduga telah membunuh ayahnya. Selain Harry, Peter juga harus menghadapi Erick Brock (Topher Grace) fotografer freelance yang mengincar pekerjaannya di harian The Bulge. Spiderman juga harus melindungi kota New York dari serbuan penjahat Sand Man Flint Marko (Thomas Haden Church). Sand dapat berubah bentuk menjadi raksasa, seperti Hulk dari pasir, tapi kuat dan tidak dapat dihancurkan lantaran dapat kembali membentuk dirinya. Dan, seakan-akan komplikasi itu tidak cukup, sang jagoan Spiderman juga harus menghadapi masalah yang timbul dari hubungan cintanya. Karir Mary Jane sebagai pemain teater hancur dan membutuhkan perhatian Peter Parker maupun sosoknya seba gai Spiderman. Spiderman yang sedang dirasuki mahkluk angkasa luar yang membuatnya jadi agresif dan sibuk mengejar Sand Man yang membunuh Ben (Cliff Robertson). Tidak ada waktu untuk Mary Jane yang sedang merajuk mencari perhatian. Sisi gelap sang Spiderman itu ditampilkan dengan berubahnya kostum laba-laba merah biru menjadi hitam setelah makhluk dari angkasa luar itu merasukinya. Namun pada menyadari akan dirinya yang telah berubah, dan Peter mencoba mengeluarkan mahluk tersebut dari tubuhnya, namun setelah mahluk itu keluar, hal yang muncul lebih buruk lagi, mahluk asing tersebut masuk kedalam tubuh Erick Brock dan akhirnya berubah menjadi Venom. Flint Marko (Sandman) akhirnya bekerja sama dengan Erick ( Venom ) untuk menghancurkan Spiderman, karena mengetahui Mary Jane merupakan pacar Peter, akhirnya Sandman dan Venom menculik Mary Jane untuk memancing keluar Spiderman, akhirnya Spiderman dihadapkan dengan dua musuh yang kuat yaitu Sandman dan Venom, setelah beberapa saat akhirnya datanglah Little Goblin Harry yang membantu Spiderman melawan Sandman dan Venom,terjadi pertarungan hebat diantara keempatnya, pada akhirnya Little Goblin meninggal tertusuk Besi akibat menolong Peter, dan Erick Brock meninggal terkena bom akibat tidak ingin melepaskan mahluk asing dari tubuhnya.
Film ini tidak hanya menceritakan tentang kisah romantis antara Peter dan Mary Jane tetapi juga menceritakan tentang persahabatan antara Peter dan Harry Osborn bagaimana mereka bisa menjaga persahabatan sampai akhir walaupun persahabatan mereka sempat terpecah.
Akting dan kostum dan make up yang digunakan dalam film ini sangat hebat dan menakjubkan, jadi meskipun semuanya hanya berupa cerita khayalan tetapi semuanya dirangkai sehingga tampak secara nyata. Hal ini lah yang membuat kita semakin penasaran untuk menonton film tersebut.
Overall, this movie berisi cerita yang sangat menarik, bukan hanya tentang kisah cinta, tetapi juga tentang persahabatan dan kekeluargaan. melalui film ini kita dapat melihat bahwa setiap orang bebas berkreasi, sperti dalam film ini meskipun hanya cerita yang khayalan namun karena proses pembuatannya yang sangat luar biasa maka menarik banyak perhatian setiap orang untuk menonton film ini.
