Uji aktivitas antibakteri dapat dilakukan dengan metode difusi dan metode pengenceran. Disc diffusion test atau uji difusi disk dilakukan dengan mengukur diameter zona bening (clear zone) yang merupakan petunjuk adanya respon penghambatan pertumbuhan bakteri oleh suatu senyawa antibakteri dalam ekstrak. Syarat jumlah bakteri untuk uji kepekaan/sensitivitas yaitu 105-108 CFU/mL (Hermawan dkk., 2007).
