Kemudian disiapkan ose yang telah steril kemudian , ambil kapang dan khamir menggunakan ose , setelah itu goreskan pada media agar miring PDA. dilakukan didepan api bunsen agar tetap sterik. kemudian diinkubasi dengan suhu 30oC selama 1-2 hari. Disimpan pada suhu rendah dan amati.
