Kegiatan kemanusiaan Tzu Chi diawali dari 6 murid Master Cheng Yen yang setiap hari merajut tambahan sepasang sepatu bayi untuk mengumpulkan dana kemanusiaan. Di samping itu, Master Cheng Yen memberi celengan bambu pada 30 ibu rumah tangga yang menjadi pengikutnya. Dia meminta agar setiap hari mereka menghemat 50 sen uang belanja dan memasukkannya ke dalam celengan itu. Dari semua hasil ini, tiap bulan terkumpul NT$ 1.170 untuk membantu kaum miskin. Kabar ini dengan cepat tersebar di Hualien, dan semakin banyak orang yang ingin bergabung. Untuk menghimpun semua kebajikan itu, Badan Bakti Amal Tzu Chi dibentuk pada bulan Mei 1966 dan berpusat di Vihara Pu Ming. Seiring waktu, badan amal ini diubah namanya menjadi Yayasan Kemanusiaan Buddha Tzu Chi.
Pada awal masa pembentukan Yayasan Kemanusiaan Buddha Tzu Chi, Master Cheng Yen bersama para pengikut mengambil tempat sempit yang tidak lebih dari 20 m² di Vihara Pu Ming, sambil berupaya menghasilkan produk untuk mendukung kehidupan, sambil mengurus jalannya organisasi. Pada musim gugur tahun 1967, ibunda Master Cheng Yen membelikannya sebidang tanah yang sekarang dimanfaatkan untuk bangunan Griya Perenungan. Walaupun demikian, Master Cheng Yen beserta para pengikut masih tetap mempertahankan prinsip hidup mandiri. Biaya perluasan seluruh proyek Griya Perenungan, selain mengandalkan pinjaman uang dari bank atas dasar hipotik hak kepemilikan tanah tersebut, juga dari hasil usaha kerajinan tangan. Sampai kini pun, Master Cheng Yen dan para pengikutnya tetap hidup mandiri dengan bercocok tanam ataupun menjalankan industri rumah tangga.
