RIANDA SRI RAHAYU SINURAT. DNA analysis of potential Barcode on Durian (Durio spp.) and Langsat (Lansium spp.) on the Farm Agroforestri in Berau, East Kalimantan. Guided by ISKANDAR z. SIREGAR.Biodiversity resources existing in Berau, East Kalimantan, such as durian and langsat is a kind of tropical fruits which are potentially for domestication. The information accurately about such types important to know in order to support the efforts of the conservation and utilization of the type appropriately. However, the introduction of this type of morphological basis still cause confusion so that it required an effort to identify the types of appropriately and accurately by DNA barcode applications. This research aims to identify the type of analysis based on the morphology of leaves, and DNA analysis of potential using barcode region matK, rbcL, and ITS. Methods undertaken, i.e., sampling the characteristic morphology of leaves, leaf, DNA extraction (CTAB), PCR, and sequencing of DNA. The results showed that the variable length of the lamina (LD), leaves (LD), widest leaf length (LP), the number of bones (JT), broad leaves (LS) and the circumference of the leaves (TOS) can be used as identifier from both types of Lansium spp., whereas in Durio spp. each variables characterizing the fifth type of Durio spp.. Locus matK and rbcL successfully diamplifikasi and disekuensing reach 100%, while ITS only able to diamplifikasi 86% and 43% disekuensing. Locus matK have averaged the longest sequence, both on Durio spp. or Lansium spp. Third locus has value to the distance smaller than the value of the intra interspesifik, but on the distribution of Kimura 2-Parameter (K2P) hasn't been able to exhibit the existence of barcoding gap. Based on a matK phylogeny locus trees capable of pegging between sample research, comparison data (GB) and data out group (OG).
