Untuk mengetahui jumlah mikroorganisme yang terdapat pada tangan dan rambut pekerja dapat dilakukan dengan cara menumbuhkan sampel yang mengandung mikroorganisme pada beberapa cawan agar. Jenis mikroorganisme yang biasanya dapat tumbuh dan diamati pada cawan agar adalah bakteri, kapang, khamir, Staphylococcus, dan jenis bakteri koliform (koliform fekal dan koliform non fekal). Uji sanitasi pekerja yang akan dilakukan saat ini adalah uji kebersihan tangan dan uji kontaminasi rambut. Uji kebersihan tangan akan dilakukan terhadap tangan sebelum dicuci, tangan setelah dicuci dengan air, tangan setelah dicuci dengan air sabun dan dibilas serta tangan setelah dicuci dengan sabun antiseptik dan dibilas. Sedangkan uji kontaminasi rambut akan dilakukan terhadap rambut yang baru dicuci dan rambut yang dicuci sehari sebelumnya (Giyarto dkk. 2004)
